Pharmacological group: Direct acting anticoagulants.
Active ingredient: Adapalene/Adapalene + Clindamycin.
Dosage form: gel.
Release form: 15ml or 30 ml in the tube.
The active ingredient in Adapalene. This is a retinoid (a substance from the vitamin A group) of the third generation. It is effective in treating mild to moderate acne, and is also used to treat keratosis pilaris and other skin conditions. Studies have shown that adapalene is as effective as other retinoids, while causing less irritation. It also has a number of advantages over other retinoids. The adapalene molecule is more chemically stable than the other two retinoids, making it suitable for use in combination with benzoyl peroxide.
Adapalene has been shown to increase the effectiveness of topical clindamycin, although side effects are also increased. Application of adapalene gel to the skin 3-5 minutes prior to clindamycin administration enhances skin penetration of clindamycin, which may improve overall treatment efficacy compared to clindamycin alone.

Klenzit has a high lipophilicity. When applied topically, it easily penetrates the hair follicles and is absorbed 5 minutes after topical application. After penetration into the follicle, adapalene binds to nuclear retinoic acid receptors. These complexes then bind to the retinoid X receptor, which induces gene transcription by binding to specific DNA regions, thus modulating the proliferation and differentiation of keratinocytes. This leads to a normalization of keratinocyte differentiation, which allows to reduce the formation of microcomedones, reduce clogging of pores and increase exfoliation by increasing cell turnover.
Adapalene is also regarded as an anti-inflammatory agent, as it inhibits the stimulated inflammatory response in the presence of Cutibacterium acnes, and inhibits both lipoxygenase activity and the oxidative metabolism of arachidonic acid to prostaglandins.
Adapalene selectively targets the retinoic acid receptor beta and retinoic acid receptor gamma when applied to epithelial cells such as skin cells. Its gamma subtype agonism is largely responsible for the observed effects of adapalene. In fact, when adapalene is used in combination with a retinoic acid receptor gamma antagonist, adapalene loses clinical efficacy.
Retination is a common temporary phenomenon reported by patients when starting to use retinols. During the initial period of treatment, the skin may become red, irritated, dry, and may burn or itch from the use of retinol; however, it tends to resolve within four weeks when used once a day.
